    Real-Time Topic and Sentiment Analysis in Human-Robot Conversation

    Socially interactive robots, especially those designed for entertainment and companionship, must be able to hold conversations with users that feel natural and engaging for humans. Two important components of such conversations include adherence to the topic of conversation and inclusion of affective expressions. Most previous approaches have concentrated on topic detection or sentiment analysis alone, and approaches that attempt to address both are limited by domain and by type of reply. This thesis presents a new approach, implemented on a humanoid robot interface, that detects the topic and sentiment of a user’s utterances from text-transcribed speech. It also generates domain-independent, topically relevant verbal replies and appropriate positive and negative emotional expressions in real time. The front end of the system is a smartphone app that functions as the robot’s face. It displays emotionally expressive eyes, transcribes verbal input as text, and synthesizes spoken replies. The back end of the system is implemented on the robot’s onboard computer. It connects with the app via Bluetooth, receives and processes the transcribed input, and returns verbal replies and sentiment scores. The back end consists of a topic-detection subsystem and a sentiment-analysis subsystem. The topic-detection subsystem uses a Latent Semantic Indexing model of a conversation corpus, followed by a search in the online database ConceptNet 5, in order to generate a topically relevant reply. The sentiment-analysis subsystem disambiguates the input words, obtains their sentiment scores from SentiWordNet, and returns the averaged sum of the scores as the overall sentiment score. The system was hypothesized to engage users more with both subsystems working together than either subsystem alone, and each subsystem alone was hypothesized to engage users more than a random control. In computational evaluations, each subsystem performed weakly but positively. In user evaluations, users reported a higher level of topical relevance and emotional appropriateness in conversations in which the subsystems were working together, and they reported higher engagement especially in conversations in which the topic-detection system was working. It is concluded that the system partially fulfills its goals, and suggestions for future work are presented

    Decline in Amphibian Health in Local Stream

    Many local streams of the Santa Monica Mountains are populated by the native California Newt, Taricha torosa, a species of special concern. Arroyo Sequit is one of these streams, the upper and lower portions of which are split by a culvert and the Mulholland Highway. This stream and the surrounding areas burned during the Woolsey fire of 2018. Since the fire, construction has been ongoing in and around the stream. Two years post-fire (during the summer of 2020) significantly more newts were found in the stream than years prior. A large proportion of these newts were unhealthy in appearance, presenting with symptoms including emaciation, cloudy eyes, infected vents, and necrotic and bleeding tails. A higher incidence of infection was noted at Lower Arroyo (below the culvert, where construction activity is concentrated) than at Upper Arroyo. Pathology studies by the National Wildlife Health Center found that these newts tested negative for ranavirus and B. salamandivorans but positive for B. dendrobatidis, a chytrid fungus to which newts are not typically susceptible. Although the final pathology report came back inconclusive, it is clear that the newt population of Arroyo Sequit is exhibiting concerning health abnormalities. We suggest that a combination of environmental stressors, including wildfires and construction, may have an immunocompromising effect. We speculate that this may be a factor in the health decline of this newt population

    Pre-service elementary social studies teachers in the southeastern United States participated in a mixed methods study to determine the degree to which they utilized critical thinking skills.  Insight Assessments administered analysis of their reflections, critical thinking skills, and dispositions test; the researchers developed a post survey for the study based on Paul & Elder’s (2009) work.  Pre-service teachers initially believed that their critical thinking abilities were above average; however, after taking the skills and dispositions test and receiving feedback on their reflections, they developed a new appreciation and understanding of the components of critical thinking. They also made a commitment to help their future students develop their critical thinking skills. This study addresses the need to consider how our pre-service teachers think, how to help them think more critically, and how to help them teach critical thinking skills when they become teachers. 

    Microstructure of Temporo-Parietal White Matter as a Basis for Reading Ability Evidence from Diffusion Tensor Magnetic Resonance Imaging

    AbstractDiffusion tensor magnetic resonance imaging (MRI) was used to study the microstructural integrity of white matter in adults with poor or normal reading ability. Subjects with reading difficulty exhibited decreased diffusion anisotropy bilaterally in temporo-parietal white matter. Axons in these regions were predominantly anterior–posterior in direction. No differences in T1-weighted MRI signal were found between poor readers and control subjects, demonstrating specificity of the group difference to the microstructural characteristics measured by diffusion tensor imaging (DTI). White matter diffusion anisotropy in the temporo-parietal region of the left hemisphere was significantly correlated with reading scores within the reading-impaired adults and within the control group. The anisotropy reflects microstructure of white matter tracts, which may contribute to reading ability by determining the strength of communication between cortical areas involved in visual, auditory, and language processing

    The incidence and mortality rates from endometrial cancer are increasing. There have been no new drugs approved for the treatment of endometrial cancer in decades. The National Cancer Institute, Gynecologic Cancer Steering Committee identified the integration of molecular and/or histologic stratification into endometrial cancer management as a top strategic priority. Based on this, they convened a group of experts to review the molecular data in this disease. Here we report on the actionable opportunities and therapeutic directions identified for incorporation into future clinical trials

    Tumor innate immunity primed by specific interferon-stimulated endogenous retroviruses.

    Mesenchymal tumor subpopulations secrete pro-tumorigenic cytokines and promote treatment resistance1-4. This phenomenon has been implicated in chemorefractory small cell lung cancer and resistance to targeted therapies5-8, but remains incompletely defined. Here, we identify a subclass of endogenous retroviruses (ERVs) that engages innate immune signaling in these cells. Stimulated 3 prime antisense retroviral coding sequences (SPARCS) are oriented inversely in 3' untranslated regions of specific genes enriched for regulation by STAT1 and EZH2. Derepression of these loci results in double-stranded RNA generation following IFN-γ exposure due to bi-directional transcription from the STAT1-activated gene promoter and the 5' long terminal repeat of the antisense ERV. Engagement of MAVS and STING activates downstream TBK1, IRF3, and STAT1 signaling, sustaining a positive feedback loop. SPARCS induction in human tumors is tightly associated with major histocompatibility complex class 1 expression, mesenchymal markers, and downregulation of chromatin modifying enzymes, including EZH2. Analysis of cell lines with high inducible SPARCS expression reveals strong association with an AXL/MET-positive mesenchymal cell state. While SPARCS-high tumors are immune infiltrated, they also exhibit multiple features of an immune-suppressed microenviroment. Together, these data unveil a subclass of ERVs whose derepression triggers pathologic innate immune signaling in cancer, with important implications for cancer immunotherapy

    A large scale hearing loss screen reveals an extensive unexplored genetic landscape for auditory dysfunction

    The developmental and physiological complexity of the auditory system is likely reflected in the underlying set of genes involved in auditory function. In humans, over 150 non-syndromic loci have been identified, and there are more than 400 human genetic syndromes with a hearing loss component. Over 100 non-syndromic hearing loss genes have been identified in mouse and human, but we remain ignorant of the full extent of the genetic landscape involved in auditory dysfunction. As part of the International Mouse Phenotyping Consortium, we undertook a hearing loss screen in a cohort of 3006 mouse knockout strains. In total, we identify 67 candidate hearing loss genes. We detect known hearing loss genes, but the vast majority, 52, of the candidate genes were novel. Our analysis reveals a large and unexplored genetic landscape involved with auditory function

    60°N was +2.0°C, relative to the 1981–2010 average value (Fig. 5.1). This marks a new high for the record. The average annual surface air temperature (SAT) anomaly for 2016 for land stations north of starting in 1900, and is a significant increase over the previous highest value of +1.2°C, which was observed in 2007, 2011, and 2015. Average global annual temperatures also showed record values in 2015 and 2016. Currently, the Arctic is warming at more than twice the rate of lower latitudes
